Protocol for short-term in vitro propagation of patient-derived group 4 medulloblastoma MBT375 tumor cells

Abstract

Durable in vitro cell culturing is an urgent unmet need for the pre-clinical development of novel group 4 medulloblastoma (G4-MB) specific therapies. Here, we present a protocol for the establishment and propagation of G4-MB tumor cells for short-term in vitro culturing. We describe brain tumor cell isolation, murine intracranial engraftment, in vitro culturing. and assay seeding. Our approach enables short-term in vitro passaging and drug screening. For long-term propagation of these G4-MB models, serial engraftment is required. For complete details on the use and execution of this protocol, please refer to Custers et al.1.
