A rapid and simple method for assaying 3H-spiroperidol binding to solubilized dopamine receptors

Abstract

Solubilized bovine caudate tissue was shown to contain specific 3H-spiroperidol binding activity indicative of dopamine receptors. These data were obtained with the aid of a hydroxyapatite batch procedure for separating bound and free ligand which proved to be rapid, inexpensive and reproducible. The specific binding thus obtained averaged between 75 and 82% of the total binding to the tissue preparation and exhibited binding characteristics (KD, drug competition order of potency) consistent with data published for particulate receptors.
